    摘要:

    目的 探讨参萸养心胶囊抗心肌纤维化的作用机制。 方法 利用TCMSP、Genecards、String、Uniprot、DAVID等数据库进行网络药理学分析,通过Cytoscape3.72软件作图及相关插件进行拓扑分析,AutoDock等软件进行分子对接。 结果 筛选出参萸养心胶囊102种成分和161个靶点,心肌纤维化靶点3 116个,疾病和药物的交集靶点43个,GO和KEGG富集分析得到174和74个条目。参萸养心胶囊治疗心肌纤维化重要成分为木犀草素、山奈酚、β-谷甾醇、丹参酮IIA、人参皂甙Rh2,重要靶点为TNF、IL6、IL1B、VEGFA、STAT3,重要生物学过程和通路为TNF信号通路、HIF-1信号通路、MAPK信号通路、RNA聚合酶II启动子转录的正调控、基因表达正调控。关键成分和关键靶点分子对接结果显示具有强烈的结合力。 结论 本研究预测了参萸养心胶囊治疗心肌纤维化的可能作用机制,为临床应用提供了理论依据。

    Abstract:

    Objective To explore the mechanism of Shenyu Yangxin Capsule in the treatment of cardiac fibrosis. Methods TCMSP,Genecards,String,UniProt,DAVID databases were used for network pharmacology analysis. Topological analysis was carried out with Cytoscape3.72. AutoDock was used for molecular docking. Results 102 ingredients and 161 targets of Shenyu Yangxin capsule were screened out. 3 116 targets of myocardial fibrosis and 43 intersection targets were screened out. 174 and 74 entries were obtained by GO and KEGG enrichment analysis. The important components of Shenyu Yangxin capsules for the treatment of cardiac fibrosis are luteolin,kaempferol β- Sitosterol,tanshinone IIA,ginsenoside Rh2. The important targets were TNF,IL6,IL1B,VEGFA,STAT3. The important biological processes and pathways were TNF signaling pathway,HIF-1 signaling pathway,MAPK signaling pathway,positive regulation of RNA polymerase II promoter transcription,positive regulation of gene expression. The molecular docking results of the key components and key targets showed strong binding ability. Conclusion This study predicted the possible mechanism of Shenyu Yangxin Capsule in the treatment of myocardial fibrosis and provided a theoretical basis for its clinical application.
